RESUMO

Essential oils (EOs) are mixtures of volatile plant secondary metabolites and have been exploited by humans for thousands of years for various purposes because of their many bioactivities. In this study, the EO from Thymus capitatus, a thyme species organically cultured on the Greek Island of Lemnos, was analyzed for its chemical composition (through GC-FID and GC-MS), antioxidant activity (AA), and total phenolic content (TPC), as well as its antimicrobial and antibiofilm actions against three important foodborne bacterial pathogens (Salmonella enterica ser. Typhimurium, Listeria monocytogenes, and Yersinia enterocolitica). For the latter investigations, the minimum inhibitory concentrations (MICs) and minimum biofilm inhibitory concentrations (MBICs) of the EO against the planktonic and biofilm growth of each pathogen were determined, together with the minimum biofilm eradication concentrations (MBECs). Results revealed that T. capitatus EO was rich in thymol, p-cymene, and carvacrol, presenting high AA and TPC (144.66 µmol TroloxTM equivalents and 231.32 mg gallic acid equivalents per g of EO, respectively), while its MICs and MBICs ranged from 0.03% to 0.06% v/v and 0.03% to 0.13% v/v, respectively, depending on the target pathogen. The EO was able to fully destroy preformed (mature) biofilms of all three pathogenic species upon application for 15 min, with MBECs ranging from 2.00 to 6.25% v/v. Overall, the results demonstrate that the EO of organically cultured T. capitatus presents strong antioxidant, antibacterial, and antibiofilm properties and could, therefore, be further exploited as a functional and antimicrobial natural formulation for food and health applications.

PURPOSE: To report an unusual case of multifocal bacterial keratitis that despite success-ful treatment caused chronic ocular hypertension. METHODS: A 67-year-old woman with unilateral multifocal keratitis and no previous ocular pathology was admitted to our hospital. Corneal scrapings and conjunctival samples were obtained for culture and the patient received intensive therapy with fortified vancomycin and tobramycin eye drops. RESULTS: The cultures demonstrated two strains of Staphylococcus epidermidis, one resistant to ciprofloxacin and both sensitive to vancomycin. Treatment was effective and gradually discontinued after total cessation of the inflammatory activity. During the follow-up period, the patient developed late and persistent ocular hypertension of unknown etiology, in absence of any detectable inflammation or complication, and received permanent antiglaucoma therapy. CONCLUSION: Differential diagnosis between fungal and bacterial infection is critical in cases of multifocal keratitis. Patients with multifocal bacterial keratitis may need intraocular pressure monitoring, even after complete infection healing.

Two different experimental techniques employing conservative tracers to determine exfiltration losses in a sewer system are reported. Both techniques compare the in-sewer responses measured along designated indicator and reference sections to assess the level of exfiltration although with different degrees of reliability. The experimental and interpretation errors associated with a pulse dosing technique, in which Rhodamine WT is used as the single tracer, are described. The estimation of exfiltration requires the comparison of indicator and reference peaks which need to be well resolved and reproducible in order to provide realistic results. A more consistent technique involves the continuous dosing of two different tracers with the objective of producing simultaneous downstream peak tracer concentrations. Any errors due to flow variations are eliminated and the longer measurement period allows both peak and background concentrations to be more accurately determined. Rhodamine WT, Li and Br were used in different combinations over the approximately 20 min duration of each continuous dosing experiment and were found to demonstrate conservative behaviours with measured recoveries of 97.6-100.6%, 97.3-101.4% and 108.4-109.2%, respectively. The analytical problems associated with the determination of the different tracers in sewage are described and are shown to be responsible for the reduced accuracy of the results associated with the use of Br. The preferred combination of tracers is Rhodamine WT as the indicator and Li as the reference and these are shown to be capable of estimating sewer losses to an accuracy of +/-1.0% using the continuous dosing approach.
